Note 1:   Despite the desire of many participants to have a test for HT35 similar to that performed by Cinnioglu, et.al., there are problems;  see the following quote from a
message received from Thomas Krahn of DNA Fingerprint:  "The 49a,b TaqI RFLP test originally requires pouring a polyacrylamide gel, blotting the DNA fragments on a nylon
membrane and hybridizing them with radioactive oligonucleotide probes. You can imagine that there is much wet chemistry for quite a little effort. Probably the radioactive
probes can be replaced by fluorescent probes (if the laboratory has a fluorescent membrane scanner), but the polyacrylamide gel is still quite toxic and the high
concentration of genomic DNA needed can only be extracted from blood samples. Possibly there is a way to amplify only 49a or 49b by PCR, but this test still has to be
designed and should be validated in comparison to the original 49a,b TaqI RFLP test. This is very much work for quite little information. I don't know any laboratory
that offers this test. As far as I know EA has also given up the idea to offer this test."  Because of the problems involved, for now, some interested participants have opted for deep
SNP testing and autosomal STR testing for special markers that might offer some insights.   This page is designed to enhance presentation of the special testing done for certain
R1b members of the DYS393=12 project, as well as comparative results received for certain individuals who have DYS393=13 or DYS393=14.
It is now apparent that not all DYS393=12 participants are ht35.   Some will be ht15, and will be associated with various  R sub-haplogroups.

A newer version of the chart may be accessed by FTDNA participants through the New Y-DNA Haplotree tab of their personal page.
Clicking on the orange "My Haplotree" tab, one willl see an "R" Haplotree showing what FTDNA believes one's test status to be.

Per Jim Elliott: "Web postings on SRY10831.2 tend to be a little confusing.  From what I've read, it seems that the SRY10831.2 is a SNP that differentiates R1a from  /|\
other R1 variants by its absence, not its presence - in other words, SRY10831.2-- defines R1a. [e.g., a positive finding, +, indicates R1b, NOT R1a]  I don't know why it
has been included in an R1b DeepSNP test.  Does everyone who has been DeepSNP-tested for R1b have a result of SRY10831.2+?  If so, it might not mean that
much." "Here are some websites discussing the SNP:
    https://lists.rootsweb.com/hyperkitty/th/read/GENEALOGY-DNA/2006-01/1136955255
See: Ann Turner, Rootsweb DNA Genealogy List Administrator:
"R1a still exists, but the marker used to determine it, SRY10831.2, is not included in  FTDNA's deep clade test. You'd have to ask them why, but I'll speculate
anyway :) A very large percentage of R1a will turn out to be R1a1 -- some  list members might have the numbers at their fingertips -- so testing M17 is
just about as good as testing both markers. Also, SRY10831.2 has some tricky interpretation issues."
    http://freepages.genealogy.rootsweb.com/~grantdna/haplogroups.html
    http://www.google.com/search?q=SRY10831.2-&hl=en&lr=&start=10&sa=N
The majority of the sites do suggest that SRY10831.2 is negative for R1a.    I do agree with you that posting SRY10831.2 results for R1b DeepSNP tests seems rather odd."

Per Kenny, Y34QR, : "I asked FTDNA why if I tested positive at SRY10831.2, I was classified as R1b1c rather than R1a. This is the answer I received 6/6/06:
"SRY10831 is the only SNP for which a negative result means that you belong to the haplogroup.  This is because it happened twice, once when haplogroups B through
R branched from A (people who are SRY10831.1 negative are in haplogroup A); and then switched back again to form R1a (SRY10831.2 negative means R1a).  This
means that everyone not in haplogroup A or R1a has a positive result for SRY10831.2.
The test you were submitted for is the Deep R1a Clade test.  We added a couple of R1b SNPs to this test panel because the Deep R1b Clade test was not ready (and
we did not know when it would be).  People who ordered the R1b SNP test, which tests only for R1b and R1b1, were switched to the Deep R1a Clade test because it
included the SNP for R1b1c.  This is why you were tested both for the R1a Deep Clades and R1b, R1b1, and R1b1c. ""

"Another useful feature of DYS464 is that the compound haplotype usually gives a good indication of the haplogroup. The data for DYF399S1 is consistent with different
haplogroups also having characteristic patterns for this marker. Curiously in both markers the highest values are found in haplogroup R. This may be coincidence or it
may indicate something about the evolutionary history of this haplogroup (or the subgroup from which the samples came, probably R1b as both the tested sample and

Note 6:  Per a 6 March 2008 message from Vince Vizachero:  "Over the past few months, a lot of progress has been made in the ability to predict ht15/ht35      <====   SEE NOTES  re testing of DYF385 & DYF411 for ht35       <====   SEE NOTES  re testing of DYF385 & DYF411 for ht35
  .... As you know, the modal value for DYS393 in ht35 is 12.  Unfortunately, many ht35 haplotypes have DYS393=13 and many ht15 haplotypes have DYS393=12 -    /|\ /|\ /|\
mostly by virtue of their sheer numbers.   DYS461=11 has always been a useful partner test, but it has never been clear how we should evaluate
a conflict between DYS393 and DYS461.  It turns out that there is a third marker, DYF385, that is particularly diagnostic
for ht35. This is not the same as DYS385, but rather a palindromic marker offered by FTDNA in its Advanced Orders. It appears that the modal value for ht15 is    /|\ /|\ /|\
DYF385=10,11 but that the modal value for ht35 is DYF385=10,10.  In fact, so far 100% of the suspected ht35 haplotypes I've tested have DYF385=10,11.
it does not appear that many members of the Border Reivers have tested either DYS461 or DYF385.  Such testing on even a small group of your members might answer
the ht15/ht35 question rather definitively… If you have members that are willing to order those two markers, I invite them to join the ht35 project so that we can
establish the probability that they truly are ht35.  [Presently], The total cost of the tests would be $28.70 (less if they had already ordered advanced tests in the  /|\ /|\ /|\
past).   See the ht 35 project at:    http://www.familytreedna.com/public/ht35/
The following is quoted from the goals statement of the ht35 project: "Our goal is to investigate the relationship between YCC Haplogroup R1b1c and
TaqI 49a,f haplotype 35 (aka ht35).  So far, no SNP has been found to distinguish ht35 from other forms of R1b1c (including the western European ht15).
Our first working hypothesis is that all ht35 haplotypes will be R1b1c* (i.e. negative for known all SNPs known to define subclades of R1b1c).
Our second working hypothesis is that ht35 contains much higher frequencies of DYS393=12 and DYS461=11 than ht15.
Our third working hypothesis is that ht35 contains much higher frequencies of DYF385=10,10 than ht15.     /|\ /|\ /|\
Therefore, our present focus is on accumulating extended Y-SNP and Y-STR testing on any R1b1c* haplotypes exhibiting DYS393=12, DYS461=11 and/or
DYF385=10,10 with the hope of finding reliable ways of distinguishing ht35 from ht15 without the need to depend on the expensive, unpleasant and
(at the moment) commercially unavailable TaqI 49a,f test."   One can hope that further research interest and a narrowing of focus will lead to definitive
  TaqI 49a,f testing !

ht35, the parent haplotype of ht25 [sic,sb "ht15"?], is most commonly found in southeastern Europe and southwestern Asia. It most likely represents a post-glacial
expansion in the Near East. Elevated levels of ht35 have also been observed among Ashkenzi and Sephardic Jewish populations.
Recently, several SNPs have been discovered that are believed to effectively demarcate ht15 from ht35. These include rs9786076 (aka S127 or L11),
   rs9785659 (aka P311), rs9786283 (aka P310), and rs34276300 (aka P312). We believe that all ht35 will be negative for both rs3427630/P312 and U106/S21." Oct,2008

DNA Tribes offers a genetic ancestry analysis that measures the connections of an   /|\
autosomal STR DNA profile of an individual to nineteen world regions….     /|\ /|\ /|\
Each world region is a characterized by a distinctive pattern of allele frequencies
across the STR loci studied….   /|\
A high degree of regional substructure is observed in world populations. However,
with the exception of a few extremely isolated populations, most areas of the world are
characterized by the intersection of more than one genetic group.   /|\

See:  A description of the DNA Tribes  Global Population Database
      /|\ /|\ /|\
Native Population Match: These results are ... Top 20 matches in a database of … native
populations  that have experienced minimal movement and admixture in modern history
(roughly, the last 500 years). Individual matches do not necessary indicate recent social or
cultural affiliation with a particular ethnicity. Instead, the geographical distribution of [ones]
Native Population Match results indicates [ones] most likely deep ancestral origins.   /|\

From sample reports:  Interpretation of Native and Global Population Match Results: Native and Global
Population Match results can be interpreted in terms of their likelihood (MLI) scores and     /|\ /|\ /|\ /|\
geographical patterns. Each Native and Global Population Match is listed with a Match
Likelihood Index (MLI) score that indicates your odds of belonging to that population relative
to your odds of belonging to a generic human population   /|\
Higher scores indicate your genetic profile is strongly represented in a population, while lower
scores indicate marginal representation in a population."

Charted below, one will find extracts from individual DNA Tribes reports indicating the top three of twenty
reported MLI's for the individual's Native Population Matches.   These top three MLI's do not necessarily   /|\ /|\ /|\ /|\
indicate a likelihood that the individual's heritage is sourced from the corresponding Global Population ever,
or at any time in the previous five hundred years at a minimum.
Examination of the   regional affinities charts  may provide insight into the extent to which a given Native Population /|\ /|\ /|\ /|\
comprises a proportion of a given Global Population, and further the extent to which members of what may be a
minority population with in the greater region may also be represented in other regional populations; eg.,      /|\
this MAY be an indicator of migration patterns from an original source "homeland" or "native population".   /|\   /|\ /|\
